A fucan from the brown seaweed Spatoglossum schröederi inhibits Chinese hamster ovary cell adhesion to several extracellular matrix proteins BJMBR
Rocha,H.A.O.; Franco,C.R.C.; Trindade,E.S.; Carvalho,L.C.M.; Veiga,S.S.; Leite,E.L.; Dietrich,C.P.; Nader,H.B..
Fucans, a family of sulfated polysaccharides present in brown seaweed, have several biological activities. Their use as drugs would offer the advantage of no potential risk of contamination with viruses or particles such as prions. A fucan prepared from Spatoglossum schröederi was tested as a possible inhibitor of cell-matrix interactions using wild-type Chinese hamster ovary cells (CHO-K1) and the mutant type deficient in xylosyltransferase (CHO-745). The effect of this polymer on adhesion properties with specific extracellular matrix components was studied using several matrix proteins as substrates for cell attachment. Treatment with the polymer inhibited the adhesion of fibronectin to both CHO-K1 (2 x 10(5))()and CHO-745 (2 x 10(5) and 5 x 10(5))...

Dynamic Cr(III) uptake by Macrocystis pyrifera and Undaria pinnatifida biomasses Electron. J. Biotechnol.
Plaza Cazón,Josefina; Viera,Marisa; Donati,Edgardo.
Background: The increased industrial activity has resulted in the discharge of large amount of pollutants including non-degradable metals into the environment. Chromium is produced in several industrial processes and it can be found in the environment in two stable oxidation states, Cr(VI) and Cr(III). Cr(VI) is more hazardous due to its carcinogenic and mutagenic effects on living organisms. Although much less toxic, Cr(III) can also exert genotoxic effects under prolonged or severe exposure. It can be separated from the solution by precipitation but biosorption using brown algae seems to be an effective and sustainable treatment technique owing to its cost-effectiveness and environmental friendly characteristics. Macrocystis pyrifera and Undaria...

Enzyme Detection and Metabolic Process Tracking of Ethanol Fermentation by a Natural Alginate Fermentation Strain BABT
Zhang,Wen; Ren,Xiaohui; Bao,Liyin.
ABSTRACT Alginate is a major component of brown algae, but it cannot be utilized for ethanol fermentation by industrial microorganisms. A natural alginate degrading and ethanol producing strain was obtained in our previous research. However, the research on the ethanol metabolism process of the natural alginate fermentation strain is lacked. In this research, the key enzyme and metabolic process of ethanol fermentation were studied. Three kinds of key enzyme including alginate lyase, pyruvate dehydrogenase and ethanol dehydrogenase were determined. The enzyme activity in the metabolic process was relatively high at 60-96 h which was the most important period during the fermentation. Meanwhile the concentration change of the important substances including...

Heterofucans from Dictyota menstrualis have anticoagulant activity BJMBR
Albuquerque,I.R.L.; Queiroz,K.C.S.; Alves,L.G.; Santos,E.A.; Leite,E.L.; Rocha,H.A.O..
Fucan is a term used to denote a family of sulfated L-fucose-rich polysaccharides which are present in the extracellular matrix of brown seaweed and in the egg jelly coat of sea urchins. Plant fucans have several biological activities, including anticoagulant and antithrombotic, related to the structural and chemical composition of polysaccharides. We have extracted sulfated polysaccharides from the brown seaweed Dictyota menstrualis by proteolytic digestion, followed by separation into 5 fractions by sequential acetone precipitation. Gel electrophoresis using 0.05 M 1,3-diaminopropane-acetate buffer, pH 9.0, stained with 0.1% toluidine blue, showed the presence of sulfated polysaccharides in all fractions. The chemical analyses demonstrated that all...

Investigation of the antifouling constituents from the brown alga Sargassum muticum (Yendo) Fensholt ArchiMer
Bazes, Alexandra; Silkina, Alla; Douzenel, Philippe; Fay, Fabienne; Kervarec, Nelly; Morin, Dimitri; Berge, Jean-pascal; Bourgougnon, Nathalie.
One of the most promising alternatives to toxic heavy metal-based paints is offered by the development of antifouling coatings in which the active ingredients are compounds naturally occurring in marine organisms and operating as natural antisettlement agents. Sessile marine macroalgae are remarkably free from settlement by fouling organisms. They produce a wide variety of chemically active metabolites in their surroundings, potentially as an aid to protect themselves against other settling organisms. In this study, a dichloromethane extract from the brown seaweed Sargassum muticum was tested in situ and, after 2 months of immersion, showed less fouling organisms on paints in which the extract was included, compared to paints containing only copper after 2...

Partial characterization and anticoagulant activity of a heterofucan from the brown seaweed Padina gymnospora BJMBR
Silva,T.M.A.; Alves,L.G.; Queiroz,K.C.S. de; Santos,M.G.L.; Marques,C.T.; Chavante,S.F.; Rocha,H.A.O.; Leite,E.L..
The brown algae Padina gymnospora contain different fucans. Powdered algae were submitted to proteolysis with the proteolytic enzyme maxataze. The first extract of the algae was constituted of polysaccharides contaminated with lipids, phenols, etc. Fractionation of the fucans with increasing concentrations of acetone produced fractions with different proportions of fucose, xylose, uronic acid, galactose, and sulfate. One of the fractions, precipitated with 50% acetone (v/v), contained an 18-kDa heterofucan (PF1), which was further purified by gel-permeation chromatography on Sephadex G-75 using 0.2 M acetic acid as eluent and characterized by agarose gel electrophoresis in 0.05 M 1,3 diaminopropane/acetate buffer at pH 9.0, methylation and nuclear magnetic...

SAMPLING FREQUENCY EVALUATION OF THE MARINE FLORA MONITORING PROGRAM AT ALMIRANTE ÁLVARO ALBERTO NUCLEAR COMPLEX, RIO DE JANEIRO, BRAZIL Boletín de Investigaciones
Garcia-Araujo,João Pedro; Hiroyuki-Kubota,Alexandre.
ABSTRACT Almirante Alvaro Alberto Nuclear Complex (CNAAA) uses seawater in the cooling system of its nuclear power plants. This water returns to the environment with a 2 °C to 8 °C temperature increase. To understand the effects of this thermal effluent, there is a large environmental monitoring program that analyses physicochemical and biological parameters, including a survey of brown algae at sampling site 32B, the nearest to the thermal discharge. This work compares monthly and seasonal variations of the specific composition of brown algae to evaluate the feasibility of changing the sampling frequency of the CNAAA monitoring program. The evaluation is performed by following two approaches: a species accumulation curve and comparisons between species...

Secondary zoospores in the algal endoparasite Maullinia ectocarpii (Plasmodiophoromycota) Biocell
Parodi,Elisa R.; Cáceres,Eduardo J.; Westermeier,Renato; Müller,Dieter G..
The present paper deals with the ultrastructure of zoospores produced by the plasmodiophorid Maullinia ectocarpii, living in the marine algal host Ectocarpus siliculosus. The zoospores described here are very similar to secondary zoospores of Polymyxa graminis and Phagomyxa sp.(the latter an algal endoparasite, also). Our results indicate that M. ectocarpii produces two types of plasmodia, and suggest that is a species with a complete life cycle, as it is known for all the Plasmodiophormycota that have been studied. Sporogenic and sporangial plasmodia produce, respectively, primary zoospores with parallel flagella within thick walled resting sporangia, and secondary zoospores with opposite flagella within thin walled sporangia.

Species Diversity, Phylogeny and Large Scale Biogeographic Patterns of the Genus Padina (Phaeophyceae, Dictyotales) ArchiMer
Silberfeld, Thomas; Bittner, Lucie; Fernandez-garcia, Cindy; Cruaud, Corinne; Rousseau, Florence; De Reviers, Bruno; Leliaert, Frederik; Payri, Claude E.; De Clerck, Olivier.
The brown algal genus Padina (Dictyotales, Phaeophyceae) is distributed worldwide in tropical and temperate seas. Global species diversity and distribution ranges, however, remain largely unknown. Species-level diversity was reassessed using DNA-based, algorithmic species delineation techniques based on cox3 and rbcL sequence data from 221 specimens collected worldwide. This resulted in estimates ranging from 39 to 61 putative species (ESUs), depending on the technique as well as the locus. We discuss the merits, potential pitfalls, and evolutionary and biogeographic significance of algorithmic species delineation. We unveil patterns whereby ESUs are in all but one case restricted to either the Atlantic or Indo-Pacific Ocean. Within ocean basins we find...
